Alanine Aminotransferase (ALT), also known as Serum Glutamic-Pyruvic Transaminase (SGPT), is a critical enzyme found predominantly in the liver. When liver cells are damaged or inflamed, ALT is released into the bloodstream, making its measurement the primary clinical indicator for liver dysfunction. Detection Principle & Scientific Methodology

The assay follows a coupled enzymatic reaction. ALT catalyzes the transfer of an amino group from L-alanine to α-ketoglutarate, generating Pyruvate and Glutamate. In the colorimetric method, pyruvate reacts with a specific probe to produce a colored product (measurable at 510–570 nm). In the UV method, the conversion of pyruvate consumes NADH, resulting in a decrease in absorbance at 340 nm. This rate of decrease is directly proportional to the ALT activity in the sample.
